Hershey-Chase experiment (1952): [Alfred Hershey and Martha Chase]

Bacteriophages: Viruses that infects bacteria. They are made-up of proteins and DNA.

1. Bacteriophage with labeled Protein: The protein coat contains 35S.

2. Bacteriophage with labeled DNA: The protein coat contains 32P.

Using above two bacteriophage two separate experiments were conducted.

In the experiments following steps took place:

Step 1: Infection. The bacteriophage is mixed with bacteria

Step 2: Blending: Bacteriophage injects genetic material inside the cell.

Step 3: Centrifugation: Separation or Bacteria and Viruses

Step 4: Analysis of radioactive P and S.

Results:

1. Bacteriophage with labeled Protein - Radioactive seen in supernatant only. Protein did not enter the cell.

2. Bacteriophage with labeled DNA - Radioactive seen in Bacteria only. DNA entered the cell.

Conclusion:

DNA is the genetic Material!!!
